PBPK Modeling of mRNA Therapeutics

COSBI is pleased to share the latest developments presented by Elio Campanile, our PhD student, who recently showcased the progress of his ongoing research as part of our latest internal talks.
Elio is developing a multiscale PBPK model that integrates key steps of mRNA-based therapeutics, including LNP-mediated delivery, cellular uptake, translation, and degradation. This model aims to improve our understanding of the biodistribution and kinetics of mRNA therapeutics in preclinical systems.
In addition, Elio presented a second project carried out in collaboration with MOLAB – Mathematical Oncology LABoratory. This work focuses on creating a multiscale mathematical model for CAR T-cell trafficking and interactions with B-cell lymphoma, combining local tumor interaction frameworks with systemic PBPK approaches. The model provides insights into CAR T-cell distribution, therapeutic efficacy, and potential strategies to support personalized treatment planning.
